目的 :研究葛根提取物對大鼠雌激素水平的影響。方法 :將 80隻成年雌性大鼠隨機分成 4組 ,每組2 0隻 :正常對照組 ;正常 +灌胃葛根提取物 ,30d ;去勢組 ,去勢 +灌胃葛根提取物 ,30d。抽血測定血清E0 ,E2 ,E3 ,睾酮等性激素水平 ;FSH ,LH ,PRL ,GH等促性激素水平。結果 :去勢後 ,E2 ,E3 顯著下降 (P <0 .0 5 ) ;E0 有所下降 ,但差異無顯著性 (P >0 .0 5 ) ,睾酮以及FSH ,LH ,PRL ,GH等垂體分泌激素增高 (P <0 .0 5 )。葛根提取物灌注後 ,正常組雌激素和促性激素水平均有不同程度的下降 ;去勢動物的E2 ,E3 恢復正常 ;E0 和睾酮比正常組增高 (P <0 .0 5 ) ,但睾酮水平與去勢組比較差異無顯著性 (P >0 .0 5 )。結論 :葛根提取物可恢復去勢大鼠的雌激素水平 ,提高促性激素水平 ;在正常大鼠體內 ,該物質表現為抗雌激素作用。 |

目的探討芹菜素在生殖軸調控中的作用及作用部位。方法通過陰道塗片選用情期成年雌性大鼠50隻,隨機分為五組,分別側腦室注入人工腦脊液、二甲基亞碸、芹菜素10-5M、hCG及芹菜素10-5M+hCG,各組再分為3d組、5d組。分別於側腦室注射後的3d、5d斷頭取血、卵巢及垂體,應用放射免疫方法分別測定血清、卵巢組織及垂體組織中FSH、LH、E2及P含量,以觀察濃度為10-5M的芹菜素對生殖軸的作用。結果1.人工腦脊液組與二甲基亞碸組無明顯差異。2.單用10-5M芹菜素組與人工腦脊液組相比FSH、LH、E2、P的含量數值小,其差異有顯著性。3.單用hCG組與人工腦脊液組相比FSH、LH的含量數值小,而E2、P的含量數值大,其差異均有顯著性。4.10-5M芹菜素與hCG合用組與單用10-5M芹菜素組相比FSH及LH的含量數值小,而E2、P的含量數值大,其差異均有顯著性。5.10-5M芹菜素與hCG合用組與單用hCG組相比FSH、LH的含量數值大,而E2、P的含量數值小,其差異均有顯著性。結論芹菜素在下丘腦水平發揮作用,進而作用於腺垂體、卵巢。10-5M芹菜素能... |

目的:評價紅花、川牛膝、丹參、女貞子、枸杞、甘草、肉蓯蓉、淫羊藿、補骨脂、菟絲子等10味中藥的植物雌激素活性。方法:選用昆明種雌性乳鼠,隨機分為A,B二組:A組又分為12組(溶劑對照組、己烯雌酚組和10個中藥組);B組也分為12組(溶劑對照組、己烯雌酚組和10個中藥拮抗組),其中中藥拮抗組每天同時灌胃中藥和己烯雌酚;各組給葯4d後分離血清。利用雌激素受體(ER)陽性MCF-7細胞,以MTT法檢測中藥含葯血清對細胞增殖的影響。結果:A組中,紅花、川牛膝、丹參、枸杞、肉蓯蓉、淫羊藿、補骨脂和菟絲子等中藥含葯血清可顯著促進MCF-7細胞增殖;而女貞子組可見細胞增殖受到明顯抑制。B組中,紅花、川牛膝、丹參、甘草、淫羊藿、補骨脂和菟絲子組可見己烯雌酚促細胞增殖的作用受到了顯著抑制;而肉蓯蓉可協同並顯著增強己烯雌酚促細胞增殖的作用。結論:紅花、川牛膝、丹參、淫羊藿、補骨脂、菟絲子等6種中藥在單獨用藥時具有擬雌激素作用,在與雌激素同時用藥時具有抗雌激素作用,其雙向調節效應依賴於體內雌激素水平的高低。 |
