這2個單詞我也不是很熟悉, 我仔細查了多個詞典, 我的理解如下

在表示蓬鬆的意思時, 二者可以互換, 比如a poofy/puffy winter jacket

而比如眼睛浮腫的, 一般用puffy eyes

而poofy則又多了一層和同性戀相關的意思
